The suburb received its name from one of the chief landowners in the area during the mid-1800s. John Jones named the suburb after Stanmore, his birthplace in England. However one of the earliest settlers in the area was Thomas Rowley, who in 1793 was given 100 acres which he named Kingston farm; this holding was later extended to 240 acres.

John Jones bequeathed 20 acres to the Methodist Church following his death in 1880, and on this site the Methodists built Newington College. Stanmore Road, which was once a dirt road, passed through the area and a tram service operated until the arrival of the railway in 1878. Stanmore Public School commenced education of the children of the area in 1884, and remains as one of the great examples of Victorian architecture in the district.
