Gregory came to Perth with his father in 1829, and commenced surveying in 1841. In 1846 he went on his first expedition north of Perth in search of pastoral land. He undertook a number of expeditions after this, including one in 1855 taking him across northern Australia from the Victoria River to Brisbane. He then became Queensland's surveyor-general, and after this went on to become a conservative politician.
